Scientific imagery of our work

Stem cell derived fetal lung organoid. Blue is dapi, green is the lung marker NKX2-1 (TTF1), red is SOX9 and Yellow is SOX2. These branching organoids make up 10-30% of the organoid cultures we get and now we can image these organoids with spectacular single cell spatial resolution.

Mouse lungs during fetal development at embryonic day 12.5. Using the Nkx2-1-IRES-mCherry reporter mouse line, the developing lung can be visualized under fluorescence microscopy showing beautifully branched structures that will continue to grow and expand reminiscent of the upside down tree. Human dF508 homozygous CF iPS-derived airway organoid after treatment with a market drug OrkambiTM resulted in swelling of the organoid and modest CFTR expression in the luminal surface of the epithelium. TTF1 (lung marker) in green, CFTR in yellow, SOX2 (proximal epithelia marker) in magenta.

Pluripotent stem cell-derived airway epithelia in air liquid interface. Green marks cilia orange marks CFTR the channel. CFTR expression on the apical membrane of ciliated cells helps mucociliary functions to clear pathogens out of the airways.
