class=\"screen-reader-text\">ORCID<\/span><\/a><\/div>[\/vc_column][vc_column width=&#8221;2\/3&#8243;][vc_column_text css_animation=&#8221;fadeIn&#8221;]<strong>Dr. Yun Li<\/strong> is a neurobiologist interested in understanding human brain development and diseases. She attended graduate school at the University of Texas Southwestern Medical Center. Her PhD work, conducted in the lab of Luis Parada, focused on using genetically engineered mouse models to study psychiatric disorders like depression and anxiety, as well as neurodegeneration such as Alzheimer\u2019s and Huntington\u2019s diseases.<\/p>\n<p>As a postdoctoral fellow, Yun worked in the lab of Rudolf Jaenisch at The Whitehead Institute and MIT. She used human pluripotent stem cells to create \u201cdisease-in-a-dish\u201d models of human brain disorders such as Rett Syndrome and microcephaly. Her work also combined brain organoid with CRISPR genome editing, to create 3-dimensional models of human brain development and diseases.<\/p>\n<p>Dr. Yun Li is currently a Senior Scientist in the Developmental, Stem Cell &amp; Cancer Biology Program at The Hospital for Sick Children, and an Assistant Professor in the Department of Molecular Genetics at the University of Toronto. I will use primary neuronal cell lines and brain organoids to validate the stability, functionality, and efficacy of the ASO-origami complexes.<\/p>\n<p><strong>Fun Facts: <\/strong>The sheer joy of being in the world of science lies in those rare moments when you hold the knowledge that the rest of the world hasn&#8217;t yet discovered, and that feeling is electrifying! Mitochondria are essential, intracellular energy-producing organelles that contain their own DNA (mtDNA). In metazoans, mtDNA is subject to high mutation rates, inherited uniparentally, and undergoes little recombination. This makes mtDNA prone to the accumulation of deleterious mutations, which can cause severe disease. To prevent deleterious mutations from being inherited, the female germline has evolved a conserved quality control mechanism, termed mtDNA selection. During this process, mitochondria undergo extensive degradation through programmed mitophagy. I apply genetic, imaging, and molecular biology techniques to study how protein BNIP3 mediates mitophagy and mtDNA selection in the female germline using fruit flies. Recent genetic studies revealed that brain somatic mutations of the PTEN-MTOR pathway are the underline causes of FCD and HME. I aim to create patient specific mutations along the pathway in human pluripotent stem cells and model these mutations in 2D neural culture and 3D brain organoids.
